I have a question about weekly pitching for Majors Division Little LEague if anyone can help. Say a kid pitches in an in house tournament game 6 innings on Monday. The tournament is played by regular season rules not tournament rules. That same team is playing in a Tournament of Champions game on Saturday which does use tournament rules. Are the pitchers that pitch 6 the week before in the in house tournament, eligable to pitch in the Tournament of champions on the same week on Saturday.

The answer is yes.

Rest period requirements are still in effect, but this is a different tournament, so the 12 year innings, and innings per week are scrubbed.

But, square it with the Tournament Director beforehand, because sometimes they don't know all the rules.

The Answer is YES. Since you are now under tournament rules, Pitching Rule 4.g. (page T-12) is in effect:
"If a player pitches in 4 or more innings in a games, 2 calendar days rest must be observed. NOTE: g. also applies between regular season games and the opening of a tournament..."

Thus, since your pitcher pitched 6 innings on Monday, he already has his 2 days rest, and can go on Saturday (which by the way is the same week).

PS this answer assumes the validity and agreement of playing a non-LL tournament game using tournament rules. Personally, I don't agree with such a decision, since it reduces participation (1 and 3 rather than 1 and 6) and puts increased strain on pitchers, but if that's what you all decided, then who is there to oppose it...?
